You can change approvers for several users at once on Settings > Users.
  1. Select the users.
  2. Select Bulk edit, then Manage approvers.
  3. Choose Add approvers, Edit approvers, or Remove approvers, then select Continue.
The dialog explains each mode. To change approvers for one user, open the Approver tab.

Who Add and Remove can update

Add approvers and Remove approvers only update users whose approver setting is Custom. Users who use a department’s managers, Microsoft profile manager sync, or No approver are skipped. Use Edit approvers to change that setting. Remove approvers also skips a user when the last custom approver would be removed. Use Edit approvers if you need No approver. A user cannot be their own approver. If some of the approvers you add are already assigned and others are not, the user is updated for those who were missing and counted as Partial. If Other departments is off in Privacy, an approver you choose must share a department with the user. Otherwise that user is skipped. Adding a second approver needs More than one approver.

Edit approvers

Edit approvers opens the same form as the Approver tab. You choose the setup. It replaces the setup for every selected user. If you choose a department, users who are not members of that department are skipped. If you choose Synchronizes the managers from the user’s Microsoft profile, each user uses the managers from their own Microsoft profile. If you choose Custom, a user is skipped when the only remaining approver would be themselves.

After you apply

absentify shows Successful, Not successful, and Partial. Partial counts users where some of the approvers you chose were already assigned. Select Finish when the run completes.
